I have a network with a mix of Windows XP and 2000 machines, plus I have a 2003 server and plan to add a second with ISA. Is it possible to change gateway IP addresses via Group Policy?

Not immediately, but you might be able to make a logon script to do this. Here is a helpful resource on how to configure TCP/IP settings from the command prompt.
